The study found that extending hairpin activators enhances CRISPR/Cas12a activation for transcription factor detection, while intact hairpins without PAM sequences are inactive, and applied this discovery to develop a CRISPR/Cas12a-based strategy for detecting NF-ĸB p50 with a linear range of 0.8 – 2000.0 pM and a LOD of 0.5 pM.
